I have VERY basic wood working skills. I need some help/assistance/advice with constructing the roof structure. Does anyone know of a good video or book that can help?

What is the proper way to seal the holes between each rafters and the interior wall? Any help or advice is greatly appreciated?

Partitions can support a roof structure, but most use trusses now-a-days. Truss design changes a little as width increases, so we need to know the width of you building. Many trusses in the 20 to 30' range use a ''W'' truss where the rafter length is divided in half and the ceiling joist in thirds with webs connecting those locations together and to the peak. Boxed soffits will close off the gaps between rafter tails.

Or with the exposed rafter tail style, bird blocks were fitted between the rafters at the outer edge of the wall. These also strengthen the roof.

Watch these 2 brothers, they hand bang all the nails. They work together perfectly too. Loads of tips and tricks. After the first video, on the RH side, you will see other videos in sequence from foundation to finish. They wil answer every question you have.

Look for any video with "finnhus" in the search, these 2 brothers made an instructional video DVD. Its worth owning.
